ABSTRACT

Objective:To analyze the spatial-temporal distribution characteristics of gonorrhea in Fujian province from 2010 to 2019, and to provide scientific evidence for developing prevention and control strategies. Methods:Data on gonorrhea cases reported in counties and districts in Fujian province from 2010 to 2019 were collected from the National Notifiable Disease Report System of the Chinese Center for Disease Control and Prevention. OpenGeoda 1.12 software was used for global and local spatial autocorrelation analysis with Moran′s I as the statistic, and SaTScan 9.6 software was used for spatial-temporal cluster analysis to explore high-risk areas. Results:A total of 56 053 cases of gonorrhea were reported in Fujian province from 2010 to 2019. The reported incidence of gonorrhea slightly increased from 2010 to 2013, decreased from 2013 to 2014, and markedly increased from 2014 to 2018 with an average annual increase rate of 8.36%. However, the reported incidence sharply dropped again in 2019. Global autocorrelation analysis showed that the global Moran′s I index ranged from 0.48 to 0.65 between 2010 and 2019 (all P < 0.05), and there was global spatial autocorrelation of gonorrhea cases in counties and districts in Fujian province. Local autocorrelation analysis showed that the main spatial clustering patterns of gonorrhea cases in Fujian province were high-high, low-low and high-low clusters. In recent years, high-high clustering areas had shown an expanding trend, and were mainly distributed in Siming and Huli districts in Xiamen city, as well as in Licheng and Fengze districts, Jinjiang, Shishi and Nan′an county-level cities in Quanzhou city. Spatial-temporal clustering analysis revealed that the first-level spatial-temporal clustering areas were counties and districts in Xiamen city, as well as central urban areas, Jinjiang, Shishi and Nan′an county-level cities in Quanzhou city from January 2015 to December 2019. Conclusions:There was a spatial and temporal clustering of gonorrhea cases in Fujian province. High-high clustering areas were mainly distributed in the economically developed areas along the southeast coast of Fujian province, and had shown a gradually expanding trend in recent years. Counties and districts in Xiamen city, central urban areas and their surrounding county-level cities in Quanzhou city should be regarded as key areas for gonorrhea prevention and control.

ABSTRACT

We investigated the distribution of Chlamydia trachomatis genotypes in the outpatients visiting STD clinics in Fuzhou City ,China .Between 2013 and 2014 ,a total of 2 019 urethral or endocervical specimens from susceptible genital C .trachomatis infections were collected and detected by using real‐time PCR assay kit .The C .trachomatis‐positive specimens were subsequently amplified with nested PCR for Omp1 region and analyzed by sequencing .Genotyping was performed by BLAST similarity search and phylogenetic tree analysis .Eighty‐three of the 86 C . trachomatis‐positive specimens were suc‐cessfully sequenced .Only one specimen was identified as multiple infection .Eight different genotypes were identified .The most prevalent was F (36 .59% ) ,followed by E (24 .39% ) ,D (12 .20% ) ,G (9 .76% ) ,J (9 .76% ) ,H (3 .66% ) ,K (2 .44% ) and B (1 .22% ) .Synonymous mutation rate and non‐synonymous mutation rate among Omp1 genes of different geno‐types were calculated .All genotypes but G showed dN/dS values of<1 .Genotypes F and E were commonest .Synonymous sub‐stitution was the major pattern of variation in the process of Omp 1 gene evolution .

ABSTRACT

Objective To investigate the effects of low molecular weight polysaccharide lsolated from agaricus blazei murill (LMPAB) on adhesion and migration of tumor cell and HUVECs. Methods HT-29 cell and HUVECs proliferation was analyzed by MTT.Adhesion and transmigration monolayer HUVECs ability of HT-29 was examined by spectrophotometry and transwell chambers. Results LMPAB showed no cytotoxic effect on HT-29 cells and HUVECs. LMPAB could significantly decrease HT-29 adhesion and transmigration through HUVECs.Conclusion LMPAB exerts the anti-tumor metastasis activity via inhibition of tumor cell adhesion and migration HUVECs.

ABSTRACT

OBJECTIVEUsing heteroduplex mobility assay (HMA) to subtype human immunodeficiency virus type 1 (HIV-1) for the purpose of understanding HIV-1 subtype epidemic in Fujian province.

METHODSDNA fragments of HIV-1 env gene were amplified from peripheral blood mononuclear cell (PBMC) cocultures of HIV-1 infected individuals by nested polymerase chain reaction (PCR). Heteroduplexs were formed through hybridizing PCR products from the samples and reference plasmid. According to the mobility of heteroduplexs in polyacrylamide gel electrophoresis, HIV-1 subtype from that sample was characterized and further confirmed by nucleotide sequencing analysis.

RESULTSThirteen of 15 (86.67%) samples were successfully subtyped by HMA, except 2 failures. Subtype E and B took up 80% (12/15) and 6.67% (1/15) respectively. Results indicated a high concordance between HMA and nucleotide sequencing analysis and concordance rate was 86.67% (13/15).

CONCLUSIONSSubtype E appeared to be the major epidemic strain of HIV-1 in Fujian. HMA showed the characteristics of fastness, easiness, economic and with high specificity, and can be used in the surviellance for the epidemic strain of HIV-1.

ABSTRACT

Aim To isolate HIV-1 from the blood specimen of HIV-infected persons, and study on the features of the isolates. Methods PBMCs from 10 heparinized blood specimens of HIV-1 infected persons were collected to isolate the virus with PBMC cocultivation method. Neuramindase was added to the culture medium to raise the HIV isolation rate. The supernatant of cocultures were detected by p24 antigen capture assay and also IFA test was used to identify the isolates. Cell tropism of the HIV-1 isolates was tested byuse of H9 and MT4 cell lines, respectively. Results HIV-1 was isolated from9 of the 10 specimens, the isolation rate could reached 90%. The 9 isolates were used to infect MT4 cells, respectively, and all of them could cause cell syncytias.8 of the isolates showed only a transient infection in MT4 cells, but the isolate FJ113 could infect MT4 successively, OD values kept no changes even it passed 15 generations, therefore, it is consider as a rapid/high replicating virus. Conclusions Addition of neuramindase into T cell culture medium may useful to raise HIV isolation rate. Most of HIV-1 isolates in Fujian are M tropic.

ABSTRACT

Objective: To investigate the intelligence development of the low birth weight babies and related factors. Method: The development ability of 190 low birth weight babies was assessed with the Revised Gesell Development Schedule. The result was compared with that of normal control. Result: The development quotients (DQ) of low birth weight infants in motor, adaptive and personal/social areas at 1 and 2 years of age and in language at 2 years of age were significantly lower than that of the normal controls. The stepwise regression analysis showed that all performances in motor, adaptive, language and personal/social areas were significantly related to age, sex, head circumstance, low birth weight and feeding way of infants, and education of their parents. Conclusion: Low birth weight infants have poorer intelligence at the age of 1 and 2 years than normal controls.
